An elav promoter regulates expression of a VP16(AD)::Zip+ hemidriver (component of a 'split driver' system, contains the VP16 activation domain). The hemidriver sequence includes the Tag:NLS(SV40-largeT) nuclear localization signal. An internal ribosomal entry site (IRES) from the Ubx gene followed by DsRed(Unk) coding sequence is present downstream of the hemidriver sequence, but this is not expressed in the nervous system.

Hsim\VP16AD.elav drives expression specifically in neurons when combined with Scer\GAL4dsx-GAL4-DBD in a split-GAL4 system including mechanosensory neurons in the terminalia associated with the bristles of the clasper tooth, lateral plates and anal plates in males, and the anal and vaginal plates in females.
Hsim\VP16AD.elav drives expression pan-neuronally, recapitulating the elav expression pattern, when combined with Scer\GAL4DBD.elav in a split-GAL4 system.
Blocking synaptic vesicle exocytosis (using Ctet\TeTxLCTNT.Scer\UAS) in the split-GAL4 combination of Scer\GAL4dsx-GAL4-DBD and Hsim\VP16AD.elav.T:Zzzz\Zip+,T:SV40\nls-largeT neurons in males and females results in a significant drop (compared to genetic controls) in courtship of females by males, males completely fail to copulate and are behaviorally sterile; females are sterile, unreceptive and show no post-mating behavioral responses.
The T:Disc\RFP tag is not expressed from this construct.
When Hsim\VP16AD.elav.T:Zzzz\ZipEERRL,T:SV40\nls2 and Scer\GAL4DBD.elav.T:Zzzz\ZipRREEL are expressed together in the same cell, they function as a complete GAL4 driver.
