Expression of Inx6GD3692 under the control of Scer\GAL4GMR23E10 in adults significantly decreases daytime and nighttime sleep duration, and sleep intensity, and significantly increases responsiveness to disturbance, compared to controls; when expression is restricted, using a Gal80[ts] transgene, to the 24 h prior to 48 h testing, daytime, but not nighttime, responsiveness is significantly increased, and daytime, but not nighttime, sleep intensity is significantly decreased, with no effect on sleep duration, compared to controls.
Expression of Inx6GD3692 under the control of Scer\GAL417d, the split-GAL4 pair of Scer\GAL4DBD.R41C07 and Hsap\RELAAD.R13F02, the split-GAL4 pair of Scer\GAL4DBD.R14C08 and Hsap\RELAAD.R12C11, or Scer\GAL4Mef2.247, but not the split-GAL4 pair of Scer\GAL4DBD.R89B01 and Hsap\RELAAD.R13F02, significantly impairs visual short term memory following training with pain stimulus in adult flies, compared to controls; this effect is also seen when Inx6GD3692 is expressed during the adult stage only, under the control of Scer\GAL4Mef2.247.Switch (induced by RU486 application for three days prior to the experiment); no memory defect is seen using Scer\GAL417d when expression is restricted to a smaller subset of cells using Scer\GAL80Mef2.mb247.
Expression of inx6GD3692 in both the anterior paired lateral (APL) and dorsal paired medial (DPM) neurons in the mushroom body (using the Scer\GAL4GH146 and Scer\GAL4c316 drivers simultaneously, and in the presence of Dcr-2Scer\UAS.cDa to increase RNAi efficiency) results in impaired 3 hour (intermediate-term) memory after associative conditioning compared to controls.
Adults expressing Inx6GD3692 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.

Prolonged optogenetic activation (using Cnoc\ChR1::Csub\ChRCsChrimson.IVS.20xUAS.Venus with (red) light) of Scer\GAL4GMR23E10 cells in adults suppresses the sleep duration decrease, but not the responsiveness increase or the sleep intensity decrease caused by expression of Inx6GD3692 under the control of Scer\GAL4GMR23E10.
