Scer\GAL4Gr28b.b.4.7 drives expression in the pattern of the isoform Gr28b-RB.
Scer\GAL4Gr28b.b.4.7 drives expression in adult abdominal multidendritic neurons with with dorsal processes extending along the left-right axis, as well as in multidendritic neurons with lateral processes extending anteroposteriorly; these major processes have several protruding dendrites. Additionally, expression is driven in multidendritic neurons with ladder-like processes along the dorsal abdominal wall. Expression is also observed in projections to the abdominal ganglion. Scer\GAL4Gr28b.b.4.7 also drives expression in neurons innervating the male reproductive organs, mostly innervating the accessory gland; and in neurons innervating the female reproductive organs, mostly innervating the common oviduct, lateral oviducts, or ovaries.
Thermogenetic activation (using TrpA1UAS.(B).cKa at 30[o]C) of Scer\GAL4Gr28b.b.4.7 cells in adults results in significantly increased feeding of satiated flies, compared to controls, this is also the case when activation is restricted to a smaller set of cells using Scer\GAL80tsh-GAL80, Scer\GAL80ple.PU, Scer\GAL80Gad1.PU, Scer\GAL80ChAT.PU, or Scer\GAL80Gad1.PU and Scer\GAL80ChAT.PU, but not Scer\GAL80elav.PY; the increase in food intake due to Scer\GAL4Gr28b.b.4.7-cell activation persists for up to 3 h after activation if no food is provided between activation and testing; increased food intake is also dependent upon the composition of the food and reflects the behavior of 12 h starved control flies; there is also a significantly increased rate of proboscis extension reflex in response to sucrose, but not in the absence of food, and significantly increased preference of satiated flies for food odor, compared to controls; there is no increase in food intake or food odor attraction due to activation in starved flies, compared to controls.
Chronic (four-day) or transient optogenetic activation (using Cnoc\ChR1::Csub\ChRCsChrimson.IVS.20xUAS.Venus with orange light) of Scer\GAL4Gr28b.b.4.7 cells in adults significantly increases food intake and proboscis extension reflex in response to sucrose in satiated flies, compared to controls; the increases in food intake and proboscis extension reflex due to transient activation persist for up to 3 h after activation, but only if no food is provided between activation and testing; chronic activation also significantly increases body weight and increases fat tissue, compared to controls, but these phenotypes recover following four days without activation, concomitant with a significant decrease in food intake, compared to controls; activation with high intensity light causes paralysis and loss of postural control, this paralysis is not seen when activation is restricted to a smaller set of cells using Scer\GAL80ChAT.PU and Scer\GAL80Gad1.PU.
Inhibition (using Ctet\tetXUAS.cUa) of Scer\GAL4Gr28b.b.4.7 cells, in combination with a Gal80[ts] transgene to restrict expression to the few days prior to testing, significantly reduces food intake and proboscis extension reflex in response to sucrose of starved and satiated flies, compared to controls; reduced food intake is also observed when activation is restricted to a smaller set of cells using Scer\GAL80ple.PU, Scer\GAL80Gad1.PU, Scer\GAL80ChAT.PU, or Scer\GAL80Gad1.PU and Scer\GAL80ChAT.PU; preference of starved flies for higher sucrose concentrations is not affected, compared to controls.
Optogenetic activation (using Crei\ChR1::Vcar\ChR1::Vcar\ChR2ReaChR.UAS.Citrine with orange light) of Scer\GAL4Gr28b.b.4.7 cells, or the combined set of Scer\GAL4Gr28b.b.4.7 and Scer\GAL4Dh44.PU cells, or a subset of Scer\GAL4Gr28b.b.4.7 cells (expression restricted using Ecol\lexAGMR65C11 and Scer\GAL80IVS.WPRE.8xlexAop), in adults significantly increases food intake of satiated flies, compared to controls; activation of Scer\GAL4Gr28b.b.4.7 cells in combination with Scer\GAL4AstA.2.1, Scer\GAL4Ilp2.PU or Scer\GAL4Gr43a-GAL4 cells in adults significantly increases food intake of satiated flies, compared to controls, but this is to a significantly reduced extent compared to Scer\GAL4Gr28b.b.4.7 cells alone.
Inhibition (using Hsap\KCNJ2UAS.cUa.EGFP) of Scer\GAL4Gr28b.b.4.7 cells, or a subset of Scer\GAL4Gr28b.b.4.7 (expression restricted using Ecol\lexAGMR65C11 and Scer\GAL80IVS.WPRE.8xlexAop), in adults significantly reduces food intake of starved flies, compared to controls.
