The temperature-sensitive paralysis caused by co-expression of stepPH.Scer\UAS.T:Avic\GFP-VC and stepPH.Scer\UAS.T:Avic\GFP-VN under the control of Scer\GAL4n-syb.PS is completely suppressed if the flies are also co-expressing both Hsap\MTORFRB.Scer\UAS.T:Uuuu\Lyn11 and Hsap\FKBP1AFKBP.Scer\UAS and are fed rapamycin.

The reduced amplitude of the excitatory junctional current at the neuromuscular junction which is seen in larvae co-expressing stepPH.Scer\UAS.T:Avic\GFP-VC and stepPH.Scer\UAS.T:Avic\GFP-VN under the control of Scer\GAL4n-syb.PS is completely suppressed if the flies are also co-expressing both Hsap\MTORFRB.Scer\UAS.T:Uuuu\Lyn11 and Hsap\FKBP1AFKBP.Scer\UAS and are fed rapamycin.

stepPH.Scer\UAS.T:Avic\GFP-VC and stepPH.Scer\UAS.T:Avic\GFP-VN are designed to be a split Venus-based probe for phosphatidylinositol 3,4,5 trisphosphate expression: when both moieties are bound to phosphatidylinositol 3,4,5 trisphosphate they concentrate, and functional Venus fluorescence is visible.
