Description

Point mutation (C2317T) in the transcript that encodes the long kis isoform; this is predicted to result in a premature stop codon (FlyBase curator comment: the mutation is reported as occurring at residue Q772, but there is no Q residue at position 772 in the reference sequence, instead the change that introduces the premature stop codon has been mapped to residue Q771).

Comment:

The mutation is reported to be at aa residue Q772 (codon CAA) but there is no Gln at position 772 in the reference sequence. It was mapped to Q771.

Detailed Description

Reference

As a result of stem cell overproliferation, kis10D26/kis10D26 somatic clones in a heterozygous background contain a greater overall number of cells and significantly more intestinal stem cells, enteroendocrine cells and enterocytes in adult flies compared to controls. The ratio between enteroendocrine cells and enterocytes is unaffected.

Comments

Expressing kisUAS.L.Tag:polyHis,Tag:FLAG (but not kisUAS.S.Tag:FLAG) under the control of Scer\GAL4Tub.PU fully rescues the increased overall number of cells and of intestinal stem cells in kis10D26/kis10D26 somatic clones, while kisFLAP or expressing kisK2060R.UAS.L.Tag:polyHis,Tag:FLAG results in only a partial rescue.
