The testis specificity index was calculated from modENCODE tissue expression data by Vedelek et al., 2018 to indicate the degree of testis enrichment compared to other tissues. Scores range from -2.52 (underrepresented) to 5.2 (very high testis bias).
dj transcript is expressed in male germ cells. A djFBtr0091350:pb-XREcollacZ construct was used to distinguish dj protein and transcript expression in the male germline. EcollacZ transcript was expressed only in premeiotic cells, while the EcollacZ protein was detected only postmeiotically, in elongated sperm bundles. Similarly, a djFBtr0091350:pb-XRGFP fusion protein was detected only in individualized sperm.
In adult testis, expression is restricted to fully elongated spermatids and is associated primarily with the spermatid axonemes. In the testis, no GFP fluorescence observed in premeiotic cells.
A 32 kD dj protein was detected in Western blots of testis extracts. A djFBtr0091350:pb-XRlacZ construct was used to distinguish dj protein and transcript expression in the male germline. lacZ transcript was expressed only in premeiotic cells, while the lacZ protein was detected only postmeiotically, in elongated sperm bundles.
