The following information accompanied stocks donated to the Bloomington Stock Center by Annabel Guichard and Ethan Bier, University of California at San Diego. Ten attB constructs were made that place SARS-CoV-2 genes nsp1–nsp10 under the control of UAS sequences. The constructs have a Kozak sequence interposed between five UAS sequences and the ATG start codon to direct high-level expression. The constructs were inserted into P{CaryP}attP2 at 68A4, 3L:11070538 (R6) or P{CaryIP}su(Hw)attP5 at 2R:14 ,307,368..14,307,690 (R6). These insertions will be denoted: P{UAS-SARS-CoV-2-nsp1.EB}attP2 P{UAS-SARS-CoV-2-nsp2.EB}attP2 P{UAS-SARS-CoV-2-nsp3.EB}attP2 P{UAS-SARS-CoV-2-nsp4.EB}attP2 P{UAS-SARS-CoV-2-nsp5.EB}attP2 P{UAS-SARS-CoV-2-nsp6.EB}attP2 P{UAS-SARS-CoV-2-nsp7.EB}attP2 P{UAS-SARS-CoV-2-nsp8.EB}su(Hw)attP5 P{UAS-SARS-CoV-2-nsp9.EB}su(Hw)attP5 P{UAS-SARS-CoV-2-nsp10.EB}attP2 In addition, two similar constructs were generated to express nsp3 protein with a C857A substitution and nsp5 protein with a C145A substitution. These substitutions render these proteases catalytically inactive. These insertions will be denoted: P{UAS-SARS-CoV-2-nsp3.C857A}attP2 P{UAS-SARS-CoV-2-nsp5.C145A}attP2
