UASp regulatory sequences drive expression of the Vinc head domain lacking the D1 region (Vinc amino acid residues 257-724 are present). The protein is targeted to the mitochondria using a C-terminal Tag:Mito(actA) membrane anchor and is also tagged at the C-terminal end with TagRFP.
The expression of VincHead-ΔD1-mito.Scer\UAS.P\T.T:Equa\eqFP578-TagRFP under the control of Scer\GAL4Mef2.PU leads to no embryonic muscle defects, as compared to controls; expression under the control of Scer\GAL4Mef2.PU also does not lead to the formation of ectopic VincHead-ΔD1-mito.Scer\UAS.P\T.T:Equa\eqFP578-TagRFP-positive cytoplasmic protein aggregates in embryonic muscles, as compared to controls.
