UAS regulatory sequences drive expression of full-length cDNA sequences from a N1S isoform of par-1 with the C-terminal 37 amino acids deleted (in the linker domain), tagged at the N-terminal with mGFP6. The cDNA used for this construct is a splice variant that lacks exon 14, which encodes 38 amino acids, including the AEM domain.
Expression of par-1N1S.ΔC.Scer\UAS.T:Avic\GFP-m6 in the follicle cells, under the control of Scer\GAL4mat.αTub67C.T:Hsim\VP16 results in a penetrant oocyte polarity phenotype, illustrated by the mispositioning of the oocyte nucelus in more than half of the egg chambers.
