Scer\UAS regulatory sequences drive expression of a tandem construct containing two copies of the Aβ (amyloid β) monomer linked together by a 12 amino acid linker peptide. Both copies of Aβ are the 40 amino acid residue form.

Flies expressing Hsap\APP[T12.Aβ40.Scer\UAS] under the control of Scer\GAL4[GMR.PF] form significant quantities of large, SDS-insoluble deposits in the head. Abundant SDS-soluble oligomeric Aβ aggregates are also present. No detectable SDS-soluble oligomeric Aβ aggregates are present. Expression of Hsap\APP[T12.Aβ40.Scer\UAS] under the control of Scer\GAL4[GMR.PF] does not result in a detectable rough eye phenotype. Flies expressing one copy of Hsap\APP[T12.Aβ40.Scer\UAS] under the control of Scer\GAL4[elav-C155] do not have a significantly different life span compared to wild-type controls.
