UASt regulatory sequences drive expression of the Hsap\SPAST M1 isoform containing the amino acid replacement C448Y (a pathogenic mutation found in HSP-SPG4 hereditary spastic paraplegia patients).
Flies expressing Hsap\SPASTC448Y.Scer\UAS.M1 under the control of Scer\GAL4elav-C155 have an aberrant posture and hold their wings abnormally apart from their body. The penetrance of this phenotype is 100%.
The time taken for animals to right themselves is significantly increased compared to wild type in flies expressing Hsap\SPASTC448Y.Scer\UAS.M1 under the control of Scer\GAL4elav-C155. The flies also show significantly reduced climbing ability.
