elav promoter sequences drive expression of the Hsap\APP Aβ42 peptide, tagged at the N-terminal end with the Tag:SS(rPENK) signal sequence.
Expression of Hsap\APPAβ42.elav.Tag:SS(rPENK) causes a significant increase in miniature excitatory post-synaptic current (mEPSC) frequency, with no difference in amplitude, in primary cultures of neurons, isolated at the embryonic stage, after 5 days in vitro, compared to controls; after 9 days in vitro, mEPSC frequency is significantly decreased, with no difference in amplitude, compared to controls. Spontaneous EPSC (sESPC) frequency, is significantly reduced at 9 days in vitro, compared to controls.
