UASt regulates expression of tbc cDNA (RE30781) tagged at the C-terminus with RFP(Unk), and which carries an Arg to Ala mutation in the TBC conserved sequence IXXDXXR that has been shown in other Rab-GAPs to render the protein a catalytically inactive dominant negative.

      The expression of RabX6UASp.YFP under the control of Scer\GAL4SPARC-MI00329-GAL4 (and Dicer-2, for a more efficient RNAi) in the larval fat body induces defects in extracellular matrix material secretion (as shown by the retention of Vkg.GFP), despite of no general secretion defects (as shown by the normal patterns of secr.GFP signal).
