Description

UAS regulatory sequences drive expression of a mutant form of TBPH in which the alanine at amino acid 405 has been replaced by threonine. The construct is tagged at the N-terminal end with mRFP1 fluorescent protein.

Expression of TBPHA405T.Scer\UAS.T:Disc\RFP in the eye under the control of Scer\GAL4GMR.PF results in severe eye depigmentation and neurodegeneration at 25[o]C.

Expression of TBPHA405T.Scer\UAS.T:Disc\RFP under the control of Scer\GAL4D42 has no effect on the number of boutons in the larval neuromuscular junction. The number of synaptic vesicles is reduced compared to wild type and supernumerary satellite boutons are not present. The number of axonal branches is also comparable with wild type.

Expression of TBPHA405T.Scer\UAS.T:Disc\RFP in motor neurons under the control of Scer\GAL4D42 results in locomotor defects in third instar larvae. Crawling larvae rolled ventral side up take longer to turn back to dorsal side up.

Expression of TBPHA405T.Scer\UAS.T:Disc\RFP under the control of Scer\GAL4D42 results in a modest amount of motor neuron apoptosis in the larval ventral ganglia.
