UASt regulatory sequences drive expression of an dsRNA targeted to a 0.7kb region of peb exon 3. It consists of a 1kb genomic peb fragment (0.3kb of the second intron and 0.7kb of exon 3) followed by a 0.7kb cDNA fragment which is in inverted orientation and corresponds to the 0.7kb exon 3 genomic fragment.
The majority of flies with pebdsRNA.Scer\UAS.cPa (one copy) driven by Scer\GAL4GMR.PU die in late pupal stages, with adult escapers: escapers have glossy eyes with mottled pigmentation, no well-defined facets, most facets lack intact lenses, and the eyes also have melanized necrotic patches on the surface. Similar but milder phenotypes are seen with pebdsRNA.Scer\UAS.cPa (one copy) driven by Scer\GAL4lz-gal4, with a gradient loss of pigmentation most severe at the posterior, and the eye has facets but is somewhat smoothened compared to wild type. When pebdsRNA.Scer\UAS.cPa (one copy) is driven by Scer\GAL4sv.PJ, there is no smoothening, loss of pigmentation, or necrosis.
