The R75G12 enhancer fragment is fused upstream of the Drosophila synthetic core promoter (DSCP) followed by the GAL4(DBD)::Zip- hemidriver (component of the 'Split GAL4' system, contains the GAL4 DNA-binding domain). A Hsp70 transcriptional terminator is present.

Inhibition (using Ctet\tetXTNT-LC.UAS or Hsap\KCNJ2IVS.10xUAS.EGFP) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.R75G12 and Hsap\RELAAD.R19G02 in adults significantly reduces takeoff rates in response to looming stimuli and almost completely abolishes short-mode takeoffs, compared to controls; in the case of inhibition using Hsap\KCNJ2IVS.10xUAS.EGFP), the giant fiber neuron has significantly reduced excitatory responses to looming and reduced depolarisation in response to slow angular velocity stimuli, compared to controls, with no effect on tonic hyperpolarisation.
Optogenetic activation (using Cnoc\ChR1Cs.IVS.20xUAS.Venus with 595 nm (amber) light) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.R75G12 and Hsap\RELAAD.R19G02 in adults results in a large and rapid depolarization of the giant fiber cell, which does not occur in controls.
Inhibition (using Ctet\TeTxLCTNT.UAS, but not Hsap\KCNJ2IVS.10xUAS.EGFP) of neurons targeted by the split-GAL4 pair of Scer\GAL4DBD.R75G12 and Hsap\RELAAD.R19G02 in adults significantly reduces jump rate in response to looming stimuli, compared to controls.
