UASt regulatory sequences drive expression of the uno coding region (amplified from genomic DNA), mutated to carry two inactivating mutations within the separase cleavage consensus motif (E103A and R133A). The coding sequence is tagged at the C-terminal end with EGFP.
Expression of unonc.UAS.EGFP under the control of Scer\GAL4VP16.bam leads to defective bivalent separation during meiosis I in wild-type and unocc1/unocc1 testes when compared to controls.
