P{UAS-Pitslre.D703A} expresses Pitslre protein with a kinase-inactivating D703A amino acid substitution under UAS control.
P{UAS-Pitslre.K587M} expresses Pitslre protein with a kinase-inactivating K587M amino acid substitution under UAS control.
The D703A and K587M mutations were generated by PCR-based mutagenesis of the LD39519 cDNA obtained from the Drosophila Gene Collection stock of Norbert Perrimon. The wild-type, D703A and K587M Pitslre sequences mutants were then excised from their backbone with the Not I and Kpn I restriction enzymes and ligated into Not I-Kpn I cut pP{UAST}.
P{UAS-Pitslre.K587M}4 is a hemizygous and homozygous viable and fertile, X chromosome insertion.
P{UAS-Pitslre.D703A}1 and P{UAS-Pitslre.K587M}1 are homozygous viable and fertile, second chromosome insertions.
P{UAS-Pitslre.N}3-1, P{UAS-Pitslre.D703A}3 and P{UAS-Pitslre.K587M}3 are third chromosome insertions.
