UASp regulatory sequences control expression of a 3.6kb genomic fragment that contains the Lkb1 transcription unit, mutated to contain a modification (S535A) that prevents phosphorylation of the gene product by Pka-C1 when expressed in S2 cells. A GFP tag has been inserted at the N-terminal end of the Lkb1 open reading frame.

Expression of lkb1S535A.Scer\UAS.P\T.T:Avic\GFP under the control of Scer\GAL4mat.αTub67C.T:Hsim\VP16 partially rescues the loss of polarity in lkb14A4-2 germ line clone oocytes. lkb1S535A.Scer\UAS.P\T.T:Avic\GFP; Scer\GAL4arm.PS rescues the lethality due to lkb14A4-2/lkb14B1-11. The resulting flies have ovaries with a disorganised follicular epithelium in which most cells appear unpolarized, and loss of anterior-posterior polarisation of oocytes.
