Overexpression of Frq2[Scer\UAS.cRPa] under the control of Scer\GAL4[D42] depresses short-term synaptic plasticity in a high Ca[2+] concentration, compared to controls. Expression of Frq2[Scer\UAS.cRPa] does not change the number of active zones per synapse in 1b boutons nor the number of docked vesicles. These mutants do not show any significant difference in resting Ca[2+] level compared to controls. Overexpression of Frq1[Scer\UAS.cRPa] under the control of Scer\GAL4[D42] significantly reduces the number of synaptic boutons. Expression of Frq2[Scer\UAS.cRPa] under the control of Scer\GAL4[D42] at 25[o]C results in reduced nerve-terminal morphology.
The number of type 1s boutons at the larval neuromuscular junction is reduced compared to wild type in animals expressing Frq2Scer\UAS.cRPa under the control of Scer\GAL4D42, while the number of type 1b boutons is not significantly affected. The amplitude of the evoked excitatory junctional potential (EJP) at the neuromuscular junction is increased compared to wild type in larvae expressing Frq2Scer\UAS.cRPa under the control of Scer\GAL4elav-C155 or Scer\GAL4D42. The amplitude and frequency of spontaneous mEJPs is not affected in these flies. Quantal content of type 1b and type 1s boutons is increased in these flies compared to wild type.

Expression of Frq2[Scer\UAS.cRPa] under the control of Scer\GAL4[D42] in a cac[TS2] background at 25[o]C results in reduced nerve-terminal morphology. Expression of Frq2[Scer\UAS.cRPa] under the control of Scer\GAL4[D42] in a cac[TS2] background at 22[o]C does no change the number of synaptic boutons compared to controls. The presence of a cac[TS2] mutant background suppresses the enhanced neurotransmitter release found upon overexpression of Frq2[Scer\UAS.cRPa] under the control of Scer\GAL4[D42] at both permissive and non-permissive temperatures.
