Description

Insertion of a P{EP} element approximately 153bp upstream of lncRNA:Hsrω. Transcription of lncRNA:Hsrω is unaffected, however, there is a severe reduction in the level of transcription of the AP-2σ gene compared to controls.

Whole-eye AP-2σang-7 clones lead to severe morphological defects: small eyes with roughening and loss of bristles; severely disorganized ommatidia of uneven size and with complete loss of photoreceptor cells; and sparse interommatidial bristles. In agreement, these mutant eyes show minimal depolarization in ERGs.

Larval muscle 6/7 NMJs in AP-2σKG02457/AP-2σang-7, AP-2σang-5/AP-2σang-7 and AP-2σang-2/AP-2σang-7 transheterozygotes show significantly more and significantly smaller boutons, as compared to controls. AP-2σKG02457/AP-2σang-7 and AP-2σang-5/AP-2σang-7 NMJs also show a significant decrease in the vesicle pool and their boutons show a significant decrease in periodic microtubule loop structures (Futsch staining), as compared to controls. Electrophysiology recording at the larval NMJ of AP-2σKG02457/AP-2σang-7 and AP-2σang-5/AP-2σang-7 transheterozygotes also show significant increases in mEJP frequency and amplitude, and significant decreases in EJP amplitude and quantal content, as compared to controls; AP-2σKG02457/AP-2σang-7 and AP-2σang-5/AP-2σang-7 transheterozygotes also show a significant decline in EJP amplitude readings in larval muscles 6/7 upon high-frequency stimulation at 34[o]C conditions, even after a 4min rest period, as compared to controls.

The expression of AP-2σUAS.cCa under the control of Scer\GAL4elav-C155 rescues the larval synaptic phenotypes but not the lethality observed in AP-2σKG02457/AP-2σang-7 or AP-2σang-5/AP-2σang-7 transheterozygotes. Expression under the control of Scer\GAL4Act5C.PU rescues both lethality and larval synaptic phenotypes in AP-2σKG02457/AP-2σang-7 transheterozygotes.

The expression of AP-2σUAS.cCa under the control of Scer\GAL4ey.PU partially rescues the eye morphology and ERG defects induced by AP-2σang-7 whole-eye clones; it fully rescues the photoreceptor cell loss.
