Nucleotide substitution: G1410?.
Compared to other alleles (notably Cyp6a291R), Cyp6a291C has a single nucleotide (G1410) deleted in the coding region - this shifts the downstream reading frame and creates a premature STOP codon. This mutant would putatively produce a C-truncated protein of 483 amino acids, which would lack the Cys residue which is the absolutely conserved amino acid functioning as the fifth ligand to the heme. The Cyp6a291C allele also has a 513-bp insertion of a 17.6 element in the 3' UTR.
A solitary long terminal repeat (LTR) of the 17.6 element is located in the 3' untranslated region. The LTR contains 513bp and is flanked by the 4bp repeated sequence ATAT.
A one bp deletion causes a frameshift at amino acid 431 of Cyp6a2.
