At 18oC, hemizygous females differentiate essentially as females but with small or rudimentary ovaries. At 25oC, neither the soma not the germ cells show evidence of oogenic differentiation. The gonad resembles a small incompletely developed testis and is populated with many small undifferentiated germ cells.

Temperature shift experiments between male and female specifying temperatures at various times during larval development demonstrated that abdominal neuroblasts adopt a male or female state at the end of the first larval instar.

Male specific muscles were found in female flies bearing any combination of tra2ts1 or tra2ts2 alleles reared at the female specifying temperature, 16oC, or the male specifying temperature, 29oC.

XX flies are fertile females at 16oC, sterile females at 18oC, and pseudomale-like intersexes at 29oC. XY flies are fertile at 16oC and sterile at 18oC and 29oC.

XX tra2ts1/tra2ts2 flies shifted to the restrictive temperature immediately after eclosion have degenerating egg chambers after 10 days at 29oC. Homozygous tra2ts2 ovaries transplanted into female hosts are abnormal and degenerate, and eggs are not deposited even when the ovary is attached to wild-type gonoducts. Homozygous XX tra2ts2 or XX tra2ts1/tra2ts2 ovaries transplanted into male hosts and kept at the restrictive temperature can mature and become vitellogenic.

XX flies lacking activity of the tra2ts2 product throughout development are somatic males.
