An Hsp70 promoter drives expression of eEF1α1.
In lines E1, E2, E3 and E4 expression is detected at 37oC. Lines E5 and E6 have no detectable expression. These lines and control lines differed mainly in the size and position of the insert. Effects of position and effects of interaction of inserts with the genetic background have significant effects on mated female and virgin male lifespan at 29 and 37oC, individuals carrying the insertions live significantly longer. Effects of insert size are smaller but significant.
Flies carrying the P{hsp70-EF-1α} construct live longer at 29oC than control flies carrying the same construct without the Ef1α48D gene.
Females with enhanced expression of Ef1α48D live significantly longer and laid significantly fewer eggs early in life than do control females. This is true at both 25oC and 29oC. Enhanced expression has no impact on the life span of virgin males.
At 25[o]C or 29[o]C transformant flies have a longer lifespan than nontransformed flies depending on the P-element insertion site.
Shepherd.
Flies carrying Ef1α48Dhs.PS have been used in a study of the impact of P-elements on fitness components in Drosophila.
