Description

The mutation in MtnA3 with respect to MtnA1 is thought to reduce Cadmium binding.

Amino acid replacement: D40K.

MtnA3 shows a number of sequence changes relative to MtnA1 : two single base changes in the promoter region, amino acid replacement E40K, a single base change and an extra 49bp segment in the 3' untranslated region.

Reported amino acid change:

D40K

Comment:

Site of nucleotide substitution in mutant inferred by FlyBase based on reported amino acid change. Author statement: There are also two single base changes in the promoter region, and one in the 3' untranslated region. The 3' untranslated region is 49bp longer than in MtnA1.

Tolerance to Cadmium the same as that as for MtnA1, despite the fact that MtnA1 is expressed at three-times the level of MtnA3.

Restriction analysis of the MtnA gene from a number of Drosophila species suggests that MtnA3 is the ancestral allele, from which MtnA1 is derived. MtnA1Dp appears to be derived from MtnA1. These two changes seem to have occurred in non-African populations.

Presumed the ancestral allele, there is presumed to be a one step transition to MtnA1 and a further step to MtnA1Dp.
