Construct: An I-element (derived from the wIR3 I-element) marked with Ecol\lacZRT gene inserted in place of a 2.6kb fragment deleted from the second open reading frame. The deletion encompasses the previously identified reverse transcriptase, RNase H and 3' zinc finger domains.
The level of sterility (i.e. the level of reactivity) of 'SF' females (sterile female progeny resulting from a dysgenic cross between strongly reactive transgenic females and inducers males) decreases within the first 12-14 generations after transgenesis. The transgenic lines have not acquired the inducer phenotype, female offspring from crosses between transgenic males and reactive females have low sterility. Low reactivity in the transgenic lines is maternally transmitted and resists repetitive crosses with a strongly reactive strain.
Defective I-element that cannot transpose. The reactivity level of the transgenic lines is tightly linked to the presence of the transgene, suggesting the transgene is a determinant of the reactivity level.
