BamHI fragment containing a Dmau\mariner element in the upstream region of Dmau\w replaces the homologous BamHI fragment spanning coordinate 1.0-4.0 in w+t11.7 (coordinates from FBrf0035956).
Construct: 3.0kb BamHI fragment of w+t11.7 has been replaced by the corresponding 4.3kb BamHI fragment from Dmau\wpch containing the Dmau\wpch Dmau\mariner insertion.
Eye colour: peach colored in the absence of Dmau\mariner transposase (in an otherwise w- background). Eye colour: mosaic peach/red (>200 spots per eye) in the presence of Dmau\mariner transposase (in an otherwise w- background).
Eye colour: peach.
w::Dmau\w transformed into D.melanogaster shows no evidence of somatic or germinal instability in the absence of other mariner elements.
In the presence of a copy of P{hsp70:Mos1}, the rate of germline excision of M{peach} from P{M{peach}wpch} is 16.9% (tested in D.melanogaster). Five out of seven insertions of P{hsp70:peach} result in a statistically significant reduction in the rate of germline excision of M{peach} from P{M{peach}wpch} caused by P{hsp70:Mos1} (tested in D.melanogaster). One out of three insertions of P{hsp70:DTBZ1} results in a statistically significant reduction in the rate of germline excision of M{peach} from P{M{peach}wpch} caused by P{hsp70:Mos1} (tested in D.melanogaster). In the presence of a copy of P{hsp26-Sgs3:Mos1}, the rate of germline excision of M{peach} from P{M{peach}wpch} is 19.9% (tested in D.melanogaster). Expression of Dtei\mariner\TDTBZ1.hs using heat shock results in a small but not significant reduction in the rate of germline excision of M{peach} from P{M{peach}wpch} caused by P{hsp26-Sgs3:Mos1} (tested in D.melanogaster). Expression of Hirr\Himar1\Ths.PL using heat shock does not result in a reduction in the rate of germline excision of M{peach} from P{M{peach}wpch} caused by P{hsp26-Sgs3:Mos1} (tested in D.melanogaster). P{hs-Hirr\Himar1\T.L} does not results in a reduction in the rate of germline excision of M{peach} from P{M{peach}wpch} caused by P{hsp70:Mos1} (tested in D.melanogaster).
Assayed in D.melanogaster.
Assayed in D.melanogaster. Used to determine the activity of a number of Dmau\mariner\T alleles.
Assayed in D.melanogaster. Used to determine the activity of a number of Dmau\mariner\T alleles and a Dsim\mariner\T allele.
One copy of P{T42I}, P{E297K} or P{M1K} mutant Dmau\mariner\T reduces the amount of w::Dmau\wpch.tGa mosaicism. P{G292R} mutant transposase allows red pigmentation in several ommatidia. P{G292R}/P{hsp70:Mos1} allows an increase in w::Dmau\wpch.tGa mosaicism over the effect of P{G292R} alone but a reduction in mosaicism compared to a single copy of P{hsp70:Mos1} : negative complementation. P{E297K} also produces a small, but noticeable reduction in w::Dmau\wpch.tGa mosaicism in combination with P{hsp70:Mos1}. In the presence of a functional transposase the peach element is excised, restoring red pigment to the eyes. In the presence of P{hsp70:Mos1} the uniform peach colour changes to dark reddish-brown, even in the absence of heat shock, resulting in a mosaic red phenotype. Assayed in D.melanogaster.
