Direct FRT sequences flank the 3' portion of w+mW.hs (the five 3' exons followed by the stop codon) and the 5' portion of w+mW.Scer\FRT.hs (promoter and followed by the first exon).
P{hsFLP} P{FRT(X96)} individuals heat shocked during mid-third instar larvae show pigment only in the posterior of the eye, the mitotic program is essentially finished. The anterior remains unpigmented because the excised extrachromosomal gene is lost when cells divide. Heat shock during early pupae (by which time cell division is completed) causes almost entirely pigmented eyes. Heat shock at late pupae show fairly uniform but light pigmentation as the eyes competence to make pigment decreases at this later stage.
The w gene is turned off in the P{FRT(X96)} construct and Scer\FLP1-mediated expression produces a functional extrachromosomal w gene that can be expressed in non-dividing cells (extrachromosomal portion is lost in dividing cells).
