Expression of a 2kb EcoRI cDNA fragment of aos including the complete coding region is driven by a Hsp70 promoter.
The low viability of argosp1e20 homozygotes is rescued by two copies of argoshs.PSb in the absence of heat shock. Expression of argoshs.PSb using heat shock almost completely rescues the external morphology of the eyes of argosp1e20 homozygotes. When argoshs.PSb is expressed using heat pulses after the wandering larval stage in argosp1e20 homozygotes, only the anterior half of the eye shows normal morphology. Expression of argoshs.PSb using heat shock also rescues the ommatidial pattern of argosp1e20 homozygotes. Expression of argoshs.PSb using a single heat pulse for 1 hour at 38oC during the third larval instar in argosp1e20 homozygotes generates a region of the eye in which most ommatidia contain the normal number of 4 cone cells, although some ommatidia in this region contain fewer cone cells than normal. As the distance from the rescued region increases, the number of extra cone cells in the ommatidia gradually increases. The abnormal wing vein and maxillary palp morphology of argosp1e20 homozygotes is rescued by expression of two copies of argoshs.PSb using heat shock.
