Description

tetO regulatory sequences plus the human cytomegalovirus virus 1E promoter drive expression of 10 copies of a Hsap\HMGA1 fragment from amino acids 2-66 containing H1 and H2 hooks in head to tail orientation. A SV40 polyadenylation signal is also present.

      Tn10\tetRey.3.5.T:Hsim\VP16-mediated expression of Hsap\HMGIYMATH20.Tn10\tetO in In(1)wm4 flies results in significant derepression of the w gene and a general loss of the variegating phenotype. Tetracycline treatment inhibits Hsap\HMGIYMATH20.Tn10\tetO-induced suppression of PEV. Application of tetracycline reveals Hsap\HMGIYMATH20.Tn10\tetO expression in undifferentiated cells is required to achieve suppression of PEV. Hsap\HMGIYMATH20.Tn10\tetO suppresses cleavage by Top2 in satellite III heterochromatin of chromosome X.
