Insertion of a P-element 23bp upstream of the first exon.
Homozygotes appear normal at 20o to 22oC, but become paralysed within 3 minutes at 37o to 38oC. Full recovery is seen within 7 to 8 minutes if the flies are transferred to 20o to 22oC. At 20oC, the resting membrane potential and the amplitude and duration of evoked excitatory junctional potentials (EJPs) at the neuromuscular junction are normal in axoP12 larvae. EJPs are lost within 10 minutes if the larvae are transferred to 38oC. During this interval the threshold required to evoke EJPs increases until transmission fails completely. Compound action potentials are either lost or reduced by more than 90%, in contrast to wild-type larvae, indicating a defect in axonal conduction. The EJP is fully recovered if the larvae are returned to 20oC.
Precise excision of the P-element confirms that it causes the axoP12 phenotype.
