Expression of amino acids 1-398 of Mmus\Ppp3ca is under the control of UASt sequences. This truncation deletes the autoinhibitory and calmodulin-binding domains, making the phosphatase activity constitutive.
At the beginning of pupariation about 25% of Mmus\Ppp3caAct.Scer\UAS; Mmus\Ppp3caAct.Scer\UAS animals fail to completely evert their anterior spiracles and retain an elongated and sometimes curved larval shape, with abdominal contraction away from the pupal case. Most other animals initiate prepupal or pupal development, with the latter everting their head sacs and evaginating wing and leg discs during day 1. However, these animals often show a cleft thorax phenotype because of an incomplete fusion of the epidermis. Externally, these animals fail to progress in developmental terms by day 4 or later as no eye, cuticle, or bristle pigmentation is observed.
