GMR regulatory sequences drive expression of the complete rl coding sequence, followed by amino acids 2-147 of Scer\GAL4 (includes the DNA-binding domain), followed by residues 413-490 of Hsim\VP16 (activation domain), followed by a Tag:HSV-1-gD epitope tag. The fusion protein is tagged at the N-terminal end with a nuclear localisation signal (Tag:NLS(SV40-largeT)).

Induction of rlGMR.T:SV40\nls2,T:Scer\GAL4,T:Hsim\VP16,T:Hsim\gD in larvae for 1 hour, followed by 6 hours recovery, results in a dramatic reduction in the fraction of cells in G1, while greatly raising the fraction in S and G2/M. After 24 hours, the fraction of cells in G2/M rises, at the expense of the pool in G1 and S.
rlSem/+ flies have a rough eye phenotype, which is enhanced if the flies also express rlGMR.T:SV40\nls2,T:Scer\GAL4,T:Hsim\VP16,T:Hsim\gD.
