P{EP}Hsp22EP3583-11 insertion maps at position -57 from the Hsp22 transcription start site, between the three heat shock elements (HSEs) and the TATA box of the promoter.
Homozygous flies have a mean lifespan of 43.9 +/- 3.7 days at 25oC, which represents a decrease of 40% compared to controls. In addition, the mutant flies rapidly enter the mortality phase, with less than 90% survival occurring at day 3 +/- 0.4 in the mutant flies, compared to day 16.9 +/- 5.3 in the control flies. Homozygous flies have a mean lifespan of 11.9 +/- 2.7 days at 30oC, compared with 20.3 +/- 0.9 days for control flies, a decrease of nearly 50%. However, the survival of homozygous flies at 37oC (667.4 +/- 89.5 hours) is not statistically different from that of control flies (442.8 +/- 203.9 hours). 20 day old mutant flies have a reduced locomotor activity of 7.9 +/- 7.8% as assessed by a negative geotaxis assay compared with 37.4 +/- 7.7% for control flies.
