UASt regulatory sequences drive expression of a mutated form of smo in which a number of phosphorylation sites in the cytoplasmic tail have been changed to Ala to prevent phosphorylation (carries the amino acid replacements S658A, S659A, S660A, S680A, S683A, S687A, S690A, S693A, S740A, S741A, T742A, S743A, S746A). The coding region is tagged with GFP.
When smoIII.V.VI.Ala.Scer\UAS.T:Avic\GFP is driven by Scer\GAL4ptc-559.1 a proximal fusion is seen of the L3 and L4 longitudinal wing veins. When smoIII.V.VI.Ala.Scer\UAS.T:Avic\GFP is driven by Scer\GAL471B a proximal fusion is seen of the L3 and L4 longitudinal wing veins near the base of the wing blade.
Carried in a plasmid and transfected into S2 cells.
