Description

UASt regulatory sequences drive expression of a mutated form of brk, in which the CiM motif has been mutated from PMDLSLG to AMAAALA, the GiM motif has been mutated from FKPY to FAAA, amino acid residues 148-200 have been changed to two amino acid residues (RS). The protein is tagged at the C-terminal end with two copies of Tag:HA.

      Adults expressing brk3M.Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4en-e16E are recovered at 25-30oC. They show loss of wing tissue or a fusion of veins IV and V and loss of the posterior crossvein. Expression of brk3M.Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4C-765 at 30oC results in wings that are slightly smaller than normal and that have vein defects, including extra crossveins.
