larval neuromuscular junction & postsynaptic membrane, with Scer\GAL4Tub.PU
Males expressing gbdsRNA.Scer\UAS under the control of Scer\GAL4αTub84B.PL show a significant amount of homosexual behaviour.
Males expressing gbdsRNA.Scer\UAS under the control of Scer\GAL4repo show a significant amount of homosexual behaviour, although less than seen when the Scer\GAL4αTub84B.PL driver is used.
The neuromuscular junctions of larvae expressing gbdsRNA.Scer\UAS under the control of Scer\GAL4tub (thus producing gb RNAi) show a large increase in postsynaptic glutamate receptor number. Consistently, the mean amplitude of spontaneous excitatory junction currents (sEJCs) from the L3 muscle 6 NMJ of these larvae ia larger than that from controls, but Scer\GAL4tub>gbdsRNA.Scer\UAS larvae do not show increase in sEJC frequency compared with wild-type controls.
Transcription from the P{UAS-gb.RNAi} construct should produce gb dsRNA, resulting in dsRNA interference (RNAi) of the gb gene.
