UASt regulatory sequences drive expression of two miRNA hairpins that each consist of a mir-6-1 precursor in which the 22bp corresponding to mature mir-6-1 have been replaced by sequences perfectly complementary to ctrip. The two hairpins target different regions of the ctrip gene. The target sites in ctrip are "CG17735-1" (5'-CAACGTCGTCTTAACCATAACT-3') and "CG17735-2" (5'-CAAGCGCGTGTCATACTAAATT-3').
Free running rhythms of flies expressing CG42574miRNA.Scer\UAS under the control of Scer\GAL4tim.PE are considerably longer (approximately 27 hours) than those of controls (24 hours). The phase response curve of these flies is shifted such that a delay, rather than an advance, occurs in response to a light pulse at ZT20. However, at ZT22 an advance is seen in both control and mutant flies.
