Imprecise excision of the progenitor insertion has resulted in an 8kb deletion that removes exons 1-3 and a portion of exon 4. The deleted sequence is all specific to isoform S97.
dlg1S97-5 flies have normal climbing ability and show a normal response in an olfactory avoidance assay (tested with benzaldehyde).
Adult dlg1S97-5 homozygous flies have severe defects in phototaxis: scores in phototaxis assays are similar to those seen in flies lacking normal phototransduction.
dlg1S97-5 homozygous males when placed with a single virgin female, spend 40% less time courting than wild-type controls, and only 20% mate within 10 minutes compared to 80% of controls.
Under alternating light/dark (12 hours) conditions, circadian patterns of locomotor activity in dlg1S97-5 homozygous adults are only slightly abnormal. However, when flies are moved to permanent dark conditions circadian patterns of locomotor activity are gradually lost.
Electrophysiological readings from the neuromuscular junctions of dlg1S97-5 homozygotes show significantly increased mini excitatory junction potential amplitude and normal evoked excitatory junction potential amplitude. Together these indicate decreased quantal content.
