Expression of Hsap\VAPBScer\UAS.cCa in neurons under the control of Scer\GAL4elav-C155 causes a dramatic increase in the number of boutons with a concomitant decrease in bouton size when compared with controls. Expression of Hsap\VAPBScer\UAS.cCa in a wild-type background causes a reduction in the average mEJP amplitude. A 50% reduction in the mEJP size and about a 10% reduction in the EJP amplitude are observed. Although there is a small decrease in the amplitude of the evoked response, the decrease in quantal size allows a nearly normal post-synaptic response.

Expression of Hsap\VAPBScer\UAS.cCa in a Vap-33-1Δ448 mutant background, under the control of Scer\GAL4C164 or Scer\GAL4D42 rescues the larval lethality. Rescued flies are fertile and do not show any obvious morphological or behavioral defects.

In synapses expressing Hsap\VAPBScer\UAS.cCa under the control of Scer\GAL4C164 or Scer\GAL4D42 in a Vap-33-1Δ448 background, the number of synaptic boutons is similar to controls. Electrophysiological analysis of the same synapses shows that the EJPs and mEJPs are both similar to controls.
