UAS regulatory sequences drive expression of a C-terminal part of pros containing an expanded 223Q polyQ tract (a 20Q tract is found in the wild-type protein). The polyQ tract is flanked by 33 amino acids on the N-terminal side and by 369 amino acids on the C-terminal side. The protein is tagged at the C-terminal end with Tag:HA.
Expression of prosZzzz\CAG.223Q.Scer\UAS.T:Ivir\HA1 in the developing eye under the control of Scer\GAL4GMR.PF results in severely collapsed eyes that lack pigmentation. An accumulation of poly(Q) clumps is detected in mutant eyes.
