7 days old females carrying homozygous germline clones result in egg chambers with 8 nurse cells and no oocyte with 30% penetrance.

Homozygous single cell DL1 neuron clones mistarget to other glomeruli besides their normal DL1 target.

Homozygous adPN and lPN clones show a significant reduction in the dendritic mass compared to wild type and dendrites spill non-specifically into inappropriate glomeruli.

Homozygous projection neuron (PN) neuroblast MARCM clones labelled using Scer\GAL4GH146 show dendritic defects, with a reduction in dendrite density and dendrites that innervate incorrect glomeruli.

Homozygous single-cell "DL1" projection neuron clones show stereotyped mistargeting defects in 19/35 cases examined; the dendrites innervate the DL glomerulus, but also mistarget several additional glomeruli (VA7m, VC2, VA6, DL2d and DL5). 7/35 of the remaining DL1 single cell projection neuron clones spill their dendrites medially and the remaining 9/35 target normally.

Homozygous projection neuron MARCM clones labelled using Scer\GAL4Mz19 show sparse innervation of the VA1d glomerulus and the dendrites are incorrectly targeted to variable glomeruli such as DA3, D and DA2. 7/9 clones target lPNs normally to DA1 with wild-type dendrite densities, while 2/9 show additional partial innervation of the adjacent DL3 glomerulus.

The axons of homozygous single-cell "DL1" projection neuron clones project into the lateral horn (as occurs in wild type), but the main branches do not reach the edge of the lateral horn. In addition, the dorsal branch of the lateral horn projection is shorter than normal or absent.

The mutant dendritic phenotypes of Dcr-1LL06357 DL1 single neuron, adPN and lPN clones are not rescued or altered by expression of Dcr-2Scer\UAS.cDa under the control of Scer\GAL4GH146.

    Comments

Precise excision of the insertion reverts the projection neuron morphogenesis defects seen in homozygous clones.
