Description

Two copies of a segment of the coding region of fkh (corresponding to amino acid positions 189-435) are sequentially cloned in a head-head orientation downstream of an Hsp70 promoter, leaving a 130bp spacer between the copies.

      Heat-shocked fkhdsRNA.hs.act salivary glands exhibit a progressive loss of tissue integrity and show strong nuclear Acridine Orange staining after puparium formation. Salivary glands could occasionally be found at later time points, but the cellular structure and shape of these glands is severely compromised.

      Staining with Acridine Orange reveals no difference between fkhdsRNA.hs.act and control salivary glands before puparium formation.
