Expression of His1dsRNA.Scer\UAS.cSa under the control of Scer\GAL4da.G32 results in lethality at the late larval or early pupal stages.
Salivary gland polytene chromosomes show a change in structure compared to in wild-type in larvae expressing His1dsRNA.Scer\UAS.cSa under the control of Scer\GAL4da.G32. Most commonly there is a broadening of the chromosome arms without an obvious disruption in the banding pattern. The DNA content of mutant and wild-type chromosomes is similar, but the mutant chromosomes show a 2- to 5-fold increase in their volume. The mutant chromosomes also show dispersion of the heterochromatic chromocenter.
Expression of His1dsRNA.Scer\UAS.cSa under the control of Scer\GAL4da.G32 results in a reproducible 14bp decrease in nucleosome repeat length from 172 to 158bp in salivary gland chromatin.
