Expression of GlySHMS01279 under the control of Scer\GAL4elav.PLu improves the climbing performance of 31-day-old, but not 5-day-old flies, as compared to age-matched controls.
Expression of GlySHMS01279 under the control of Scer\GAL4Mef2.PR is able to significantly reduce the size of autophagosomes in the muscles of chloroquine-treated larvae starved on low-nutrient food for 6 hours. The number of autophagosomes is not significantly changed compared to controls. The myopathy normally seen in starved larvae treated with chloroquine is significantly reduced, with a much-improved sarcomere structure and much less distortion of the intermyofibrillar spaces.
No appreciable effect on autophagosome size is seen when GlySHMS01279 is expressed in the fat body under the control of Scer\GAL4cg.PU.
After 3 or 6 hours of starvation, expression of GlySHMS01279 under the control of Scer\GAL4Mef2.PR significantly improves the crawling defects seen in chloroquine-treated larvae compared to controls. After 6 hours starvation, crawling is negatively affected in the absence of chloroquine treatment. In well-fed larvae there is little difference between crawling time compared to controls, either in the presence or absence of chloroquine.
