Description

Recombination between the two progenitor insertions has resulted in the deletion of the sequence between them, removing exon 4 through 12 of the Ih gene. A composite PBac{PB3.WH3} element is present at the site of the deletion.

Under light-dark conditions, only 82.7% of homozygous files are rhythmic and 17.3% of are weakly rhythmic for locomotor activity, compared to 100% of control flies being rhythmic for locomotor activity. The mean overall locomotor activity of the mutant flies is lower than normal, and the peak of activity at dawn is blunted. Approximately half of the mutant flies lack the normal bimodal pattern of locomotor activity, instead having sustained activity resembling that seen in wild-type flies during the subjective day under constant dark conditions.

Under constant darkness conditions only 24.5% of homozygous flies are rhythmic for locomotor activity. The mutant flies cannot sustain the characteristic plateau at the end of the subjective day which is seen in wild-type flies under these conditions. The period length of the mutant flies shortens (to 23.3 hours) under these conditions and activity decays in the second half of the subjective day.

Under light-dark conditions, mutant flies lack the normal cyclic pattern of dopamine expression during the daytime, and show a notable increase in dopamine levels at night time compared to wild type. Under constant darkness conditions, mutant flies show an increase in dopamine levels similar to that of night under light-dark conditions.

During the day, total length of waking period in mutant flies is not different from wild type. Total sleep both during the day and night is normal in mutant flies. However, the mutant flies show a significant increase in the number of sleep episodes and a significant decrease in the mean duration of a sleep bout compared to wild type. This sleep fragmentation is seen during both the day and night, but is more pronounced during the nighttime.

Mutant males reared at 25[o]C do not show a reduction in the maximum life-span and only a slight reduction (10%) in the median, but the death rate during the exponential phase is increased compared to wild type. Mutant flies show normal resistance to oxidative stress.
