A modified orb2 genomic fragment has been inserted into the attP site present in orb2attP via phiC31-mediated recombination. The resulting allele contains an insertion of a single nucleotide in the A isoform specific exon, which disrupts the the orb2A isoform but leaves the orb2B isoform (which is tagged at the C-terminal end with GFP) intact. In addition, the Q domain has been deleted. A single mFRT11 site is present downstream of the orb2 coding sequence, and single attL and attR sites (generated during the recombination process) are also present.
Homozygous orb2ΔQΔA.T:Avic\GFP male flies have almost no long term 24 hour memory when exposed to a courtship conditioning paradigm. Unlike wild type flies, males that experience futile courtship of mated and therefore unreceptive females fail to suppress their courtship towards other mated females. Short term 1 hour memory is comparable to wild type.
orb2ΔQΔA.T:Avic\GFP/orb2ΔB.T:Avic\GFP male flies show defects in long term 24 hour memory when exposed to a courtship conditioning paradigm, and these are similar in severity to orb2ΔB.T:Avic\GFP/orb2ΔA.T:Avic\GFP males. Unlike wild type flies, males that experience futile courtship of mated and therefore unreceptive females often fail to suppress their courtship towards other mated females.
