Description

Excision of the FRT-w cassette from progenitor AxnΔPP2A.FRT.αTub84B.Tag:FLAG allows the αTub84B promoter to drive expression of an Axn deletion mutant lacking the PP2A domain (K568-P684) which is fused with N-terminal 6xTag:FLAG.

      Expression of maternally deposited AxnΔPP2A.αTub84B.T:Zzzz\FLAG reduces the hatch rate of embryos to 6-10% of controls. Examination of larval cuticles reveal the loss of 1-2 smooth cuticle bands.

      The hatch rate of embryos expressing zygotic AxnΔPP2A.αTub84B.T:Zzzz\FLAG is not significantly affected. The majority of embryos complete embryonic development. However, the majority of the animals die before reaching adulthood.

      AxnS044230 mutant (maternal-/zygotic-) embryos expressing maternally provided AxnΔPP2A.αTub84B.T:Zzzz\FLAG exhibit deletions of smooth cuticle bands and the appearance of ectopic denticles.
