UASp regulatory sequences drive expression of Cnb which is tagged with EYFP and with a Tag:PACT(Plp) sequence that targets the protein to the centrioles.
Interphase larval neuroblasts expressing CnbScer\UAS.P\T.T:Avic\GFP-EYFP,T:PACT-cp309 under the control of Scer\GAL4unspecified contain two apical centrosomes, instead of the single centrosome present in wild type. Spindle assembly in these cells is qualitatively different from that of control cells. Asymmetric mitosis is accomplished in the mutant neuroblasts, and the orientation of the plane of cell division is maintained from one cell cycle to the next.
