UASt regulatory sequences drive expression of two copies of the M13 peptide of Ggal\MLCK, followed by a nuclear localization signal (Tag:NLS(Unk)) and the Tag:MYC epitope. The encoded protein is a blocker of nuclear Ca[2+] signaling.
Flies expressing Ggal\MLCK2xM13.Scer\UAS.T:Uuuu\nls6,T:Hsap\MYC under the control of Scer\GAL4hs.PU during conditioning (expression induced by a 30 minute heat shock 12-18 hours prior to conditioning) show no significant impairment in short-term memory, middle-term memory or anesthesia-resistant memory, but show a significant impairment in long-term memory, in an olfactory avoidance conditioning assay.
